The ester-based vanadium complexes, bis(methylacetoacetato) oxovanadium (IV), bis(ethylacetoacetato) oxovanadium(IV) and bis(ethylbenzoylacetato) oxovanadium(IV), have been prepared and characterized by physicochemical and spectroscopic methods. The biological efficacies of the complexes were investigated. These complexes possess substantial glucose lowering ability and also show improved glucose tolerance effect. © 2010 Springer Science+Business Media B.V.

The physicochemical properties, thereby the biological efficacy, of metal complexes are affected based on their structure and geometry that vary in the presence of coordinating and non-coordinating solvents. To investigate this, in the present work, we synthesized three hitherto unreported β-diketone-based oxovanadium(IV) complexes, namely [VO(tfdmh)2] (tfdmh =1,1,1-trifluro-5,5-dimethyl-2,4-hexanedione), [VO(dmh)2] (dmh = 2,2-dimethyl-3,5-hexanedione) and [VO(dbm)2] (dbm = 1,3-diphenylpropane-1,3-dione), and characterized them using electron paramagnetic resonance, UV–visible, Fourier transform infrared and electrospray ionization mass spectroscopies and single-crystal X-ray diffraction. The structural changes in the presence of dichloromethane, dimethylsulfoxide and dimethylformamide were analysed using spectroscopic techniques. Further, in vitro glucose uptake efficacy and cytotoxicity were assessed using C2C12 (rat skeletal muscle) and HeLa (human cervical cancer) cell lines, respectively. Effect of solvent coordination on the structure of β-diketone-based vanadyl complexes and assessment of in vitro antidiabetic activity and cytotoxicity

Two diketone based oxovanadium complexes, viz., bis(4,4,4-trifluoro-1-phenylbutane-1,3-dionato)oxovanadium(IV) (1) and bis(1,1,1-trifluoropentane-2,4-dionato)oxovanadium(IV) (2), have been synthesized and characterized by spectroscopic and analytical techniques. The DNA binding and the cleaving ability of the complexes is assessed by UV-vis spectroscopy, fluorescence spectroscopy, viscometry and gel electrophoretic studies. The DNA binding constant values (Kb) are found to be 1.95±0.16×103M-1 for complex 1 and 1.064±0.17×103M-1 for complex 2, respectively. Based on the results of the spectral and viscosity studies, it is observed that the complexes, interestingly, have preferred minor groove binding with DNA. Further, the concentration-dependent oxidative cleavage pattern of pBR322 in the presence of the activating reagent, hydrogen peroxide, has also been discussed. In addition, the complexes have shown moderate cytotoxic activity by inducing apoptosis against the cervical cancer cell line, HeLa. The results of in silico analysis and logP predictions are found to be in good agreement with the experimental observations. Thus, synthesized oxovanadium complexes have displayed promising DNA binding behavior and DNA cleavage activity with moderately cytotoxic nature. © 2015 Elsevier B.V.

Exploration of DNA binding mode, chemical nuclease, cytotoxic and apoptotic potentials of diketone based oxovanadium(IV) complexes

Diabetes mellitus is one of the metabolic disorders resulting from excess production of reactive oxygen species (ROS) over the normal level and also due to dysfunction in the antioxidant defense mechanism, possibly both occurring simultaneously. Excess ROS are normally scavenged by antioxidants. Transition metal complexes exhibit such antioxidant property by scavenging the excess free radicals (one type of ROS) in such disorders. In this study, we report on synthesis, characterization, and in vitro study on antioxidant property of a few new ester-based vanadium(IV) complexes, namely, bis(1,1- dimethylethylpropanedioato)oxovanadium(IV) - [VO(DTBM)2]; bis(methyl-3-oxopentanoato)oxovanadium(IV) - [VO(MPA)2]; bis(ethyl-3-oxopentanoato)oxovanadium(IV) - [VO(EPA)2]; bis(2-methyl-1,3-dimethylpropanedioato)oxovanadium(IV) - [VO(DMMM)2] and bis(2-bromo-1,3-diethylpropanedioato)oxovanadium(IV) - [VO(BDEM) 2], and also furan-based vanadium complexes. Antidiabetic potential of selected complexes has been estimated. An attempt has also been made to correlate antioxidant and antidiabetic potentials of selected complexes. Our study reveals that compounds exhibiting higher glucose lowering effect seem to possess good antioxidant potential as well. © 2012 Springer Science+Business Media New York.

A possible correlation between antioxidant and antidiabetic potentials of oxovanadium(IV) complexes

We have synthesized furan-based vanadium complexes, bis(5-nitrofuran-2- carboxylato)oxovanadium(IV) - [VO(5NF)2], bis(1-furan-2-yl- ethanonato)oxovanadium(IV) sulfate - [VO(2AF)2]SO4, and bis(5-methyl-2-furalato)oxovanadium(IV) sulfate - [VO(MFFA)2]SO 4 possessing [VO(O4)] coordination mode. These complexes are characterized by physico-chemical and spectroscopic methods. Based on electron paramagnetic resonance parameters, the proposed geometry is close to a distorted square pyramid. Animal study was carried out using standard protocol and the complete profile of glucose, protein, and total cholesterol levels were analyzed followed by an oral glucose tolerance test. © 2011 Taylor &amp; Francis.

Synthesis, spectral characterization, and antidiabetic study of new furan-based vanadium(IV) complexes

Since 1985, when Heyliger et al. first reported the in vivo insulin mimetic activity of oral vanadate, extensive studies exploring vanadium chemistry, including the synthesis of novel complexes and their biological effects both in vitro and in vivo have been pursued. Such complexes have emerged as possible potential agents for diabetes therapy. Among the several existing compounds, diketone based vanadium complexes have been chosen for the current study. Two new complexes namely bisdimethylmalonatooxovanadium(IV) and bisdiethylmalonatooxovanadium(IV) have been synthesized and characterized by UV-visible, FTIR and mass spectral studies. The antidiabetic activity of the complexes was proved by animal study. The results show that the above complexes have comparable antidiabetic potential with respect to the standard drug as well as with bisacetylacetonatooxovanadium(IV) which has been studied earlier by Reul et al. © 2008 Elsevier Masson SAS.
