Abstract
The objective of this study is to screen for the α-Amylase activity of a marine actinomycete strain Streptomyces sp. VITMK1 isolated from soil sediment sample collected from the mangrove region of Pichavaram, Tamil Nadu, India. Streptomyces sp. VITMK1 was isolated from soil sediment sample collected from the mangrove region of Pichavaram. The isolate was subjected to submerged fermentation in tryptone yeast extract (ISP No.1) medium and after the optimal duration, the cell-free supernatant was collected by centrifugation and used to check α-Amylase activity by 3,5 dinitrosalicylic acid (DNSA) method. A standard maltose curve was used to estimate the amount of maltose present in the sample after the reaction. After five minutes of assay conditions, 160 µg/ml of maltose was released by the activity of α-Amylase produced by the strain Streptomyces sp. VITMK1 and the enzyme activity was calculated as 0.934 U. The results of the study revealed that actinomycetes are a potent source for extracellular α-Amylase production. By optimizing the reaction conditions the enzyme activity of α-Amylase could be substantially increased. © RJPT All right reserved.
