An efficient, three-component domino reaction of dimedone 1, aromatic aldehydes (2a-o), and 1,3-cyclohexanedione 1a in the regio-selective synthesis of 3,3-dimethyl-9-phenyl-2H-xanthene-1,8(5H,9H)-diones (3a-o) is reported. Regioselective synthesis of 3,4,6,7-tetrahydro-3,3-dimethyl-9-phenyl-2H-xanthene-1,8(5H,9H)-diones through ascorbic acid catalyzed three-component domino reaction

A facile and efficient method for the preparation of 3,4,6,7-tetrahydro-9- (1,2-dihydro-2-oxoquinolin-3-yl)-2H-xanthene-1,8(5H,9H)-diones from substituted 2-chloro-3-formylquinoline and 1,3-cyclohexanedione or dimedone in water at 90 °C using parallel synthesizer is reported. The present methodology offers a tandem-cascade methodology, high yield, and operational simplicity. © 2014 Elsevier Ltd. All rights reserved.

Water mediated catalyst-free efficient domino synthesis of 9-(quinolin-2(1H)-one)-xanthene-1,8(5H,9H)-diones using parallel synthesizer

An efficient and simple thiourea-mediated regioselective synthesis of symmetrical and unsymmetrical diversified thioethers is reported. The regioselective reaction avoids byproduct formation and offers simplified methodology, wider applicability, and easy workability and an environmentally friendly approach toward symmetrical and unsymmetrical thioethers. The mechanism of formation of thiols and symmetrical and unsymmetrical thioethers involving a sulfur surrogate is described. © 2014 American Chemical Society.

Thiourea-Mediated Regioselective Synthesis of Symmetrical and Unsymmetrical Diversified Thioethers

Methoxy-substituted chalcones, 3 were obtained using simple, efficient method from 2-naphtylethanone, 1 and aromatic aldehydes, 2. The in vitro cytotoxicity activities of the chalcones against a panel of three human cancer cell lines were explored. The tested compounds were found to possess significant cytotoxic activity. The DNA strand break and damage was quantified through alkaline comet assay, flow cytometric analysis, and chromatin condensation studies, which revealed the apoptotic nature of the compounds. Compound 3c, (3-(3,4,5-trimethoxyphenyl)-1-(2-naphthyl) prop-2-en-1-one) showed highest cytotoxicity of 0.019 μm against HeLa, 0.020 μm against HCT15 and 0.022 μm against A549. Compound 3e, (3-(3,5-dimethoxyphenyl)-1-(2-naphthyl) prop-2-en-1-one) showed better IC50 values against all the three cell lines employed for the study.
